Antecedentes y objetivos: Esta tesis forma parte del programa de mejoramiento genético en el género Nierembergia, que iniciara en 2002 en el Instituto de Floricultura de INTA. Los objetivos generales que propuse al iniciarla, fueron: (i) conocer la compatibilidad interespecífica entre las especies nativas de valor ornamental y (ii) obtener híbridos interespecíficos como fuente de variación en el mejoramiento. Material y Métodos: En primer lugar se buscó establecer el grado de cercanía entre ocho especies del género Nierembergia: N. aristata, N. browalloides, N. calycina, N. ericoides, N. micrantha, N. linariaefolia var. linariaefolia, N. scoparia, N. veitichii en un análisis filogenético con los datos obtenidos usando el gen rpl14 y los espacios intergénico (IGS) trnK y trnL, como marcadores moleculares. En estas mismas especies se realizaron cruzamientos dirigidos aplicando polinización artificial seguida del estudio de la germinación de los granos de polen sobre el estigma y el crecimiento de los tubos polínicos a lo largo de los pistilos. Esto último permitió establecer la relación Estigma-Polen/Estilo- Tubo polínico (EPET) entre todas las especies consideradas. Con el fin de estudiar las barreras post fertilización tanto en los cruzamientos compatibles como en los incompatibles, se estudió del desarrollo del endosperma y del embrión. Finalmente se caracterizaron los híbridos interespecíficos. Resultados claves y Conclusiones: El análisis filogenético reveló la formación de tres subclados N. veitichii-N. aristata, N. linariaefolia-N. micrantha y N. scoparia-N. ericoides La distancia filogenética entre las especies que integran cada uno de ellos, fue corroborada por el análisis de compatibilidad interespecífica, y por los resultados de los estudios de endospermogénesis y embriogénesis. La combinación entre N. ericoides y N. scoparia resultó compatible, presentándose como óptima para la obtención de híbridos interespecíficos. En este género, el análisis filogenético utilizando los genes el gen rpl14 y los espacios intergénico (IGS) trnK y trnL constituye una herramienta del mejoramiento que permite predecir la compatibilidad interespecífica y consecuentemente obtener híbridos estables. |

Backgrounds and aims: This study is part of the Genetic Improvement Program for the genus Nierembergia that commenced in 2002 in the Instituto de Floricultura of INTA. The general objectives that I proposed upon initiating it were: (i) determine interspecific compatibility between native species with ornamental value; and (ii) obtain interspecific hybrids as a source of variation in the improvement. Material and Methods: The first step was to establish the degree of proximity among eight species from the genus Nierembergi: N. aristata, N. browalloides, N. calycina, N. ericoides, N. micrantha, N. linariaefolia var. linariaefolia, N. scoparia, N. veitichii in a phylogenetic analysis with data obtained using the gen rpl14 and the intergenetic spacers (IGS) trnK and trnL, as molecular markers. In these species, directed crossings were carried out for both pollen germination (on the stigma) and pollen tube growth (along the pistils). The latter allowed for establishment of the relationship Stigma-Pollen/Style-Pollen tube (SPST) in all of the crossings. Endosperm and embryo development were analyzed in order to study the post-fertilization barriers in both the compatible and incompatible crossings. Finally, the characteristics of the interspecific hybrids obtained were described. Key Results and Conclusion: Phylogenetic analysis revealed the formation of three subclads: N. veitichii-N. aristata, N. linariaefolia-N. micrantha and N. scoparia-N. ericoides. The conclusions reached concerning the phylogenetic distance between the species that integrate each one of these was coincided with that from both compatibility and embryological studies. The combination between N. ericoides and N. scoparia was compatible, allowing production of interspecific hybrids. In this genus, phylogenetic analysis using the gene rpl14 and the intergenetic spacers (IGS) trnK y trnL constitutes a tool for genetic improvement that permits the prediction of interspecific compatibility and, consequently, obtainment of stable hybrids.
