The model rests on the following assumptions: (1) The active site of the enzyme molecule can exist in at least two thermodynamically stable states: open and closed; (2) only the open conformation exchange substrates with the medium; (3) the enzyme in the closed conformation acquired over its substrate the maximal catalytic activity, (4) the fluctuation of the enzyme molecule between both conformations is an absolute requirement for the completion of a catalytical cycle. The parallelism of this cyclic catalytic process with phase transfer catalysis is discussed. Positive and negative cooperativity due to the coupling of conformational fluctuation in oligomers is predicted by the model. The same is true for half of the sites reactivity.
