On the other hand, melatonin is known for its multipurpose action due to its capacity to attenuate structural and functional alterations in several organs with high functional activity and mitotic rates. In the present study, we evaluated histological changes in ducts and acini of the submandibular gland of rats treated with cyclophosphamide, as well as the protective effect of exogenously administered melatonin. Three-month-old adult male Wistar rats with body weight ranging from 300 to 350 grams, were used and divided into five experimental groups: 1) C: Control, 2) Control and Ethanol, 3) Cyclophosphamide, 4) Cyclophosphamide and Melatonin, and 5) Melatonin. Histological sections of submandibular glands were prepared and stained with Hematoxylin/Eosin, where possible structural changes in ducts and acini were analyzed. Glands from animals treated with cyclophosphamide showed acinar cells with macrokaryosis and hyperchromasia, binucleation with irregular contours, prominent nucleoli, and perinuclear chromatin clumps compared to control groups. It is inferred that melatonin administration at the tested dose could attenuate the structural alterations of the submandibular gland treated with this chemotherapeutic agent. The reversal of such changes would explain the reduction of antioxidant and functional effects at the glandular level described in our laboratory.

Trabajos previos de nuestro equipo demostraron un efecto oxidativo y funcional de esta droga evaluado a través del incremento de superóxido dismutasa, ácido úrico y niveles de glucógeno en glándula submandibular de rata. Por otro lado, se conoce la acción de melatonina como agente multipropósito por su capacidad de atenuar alteraciones en la estructura y función de varios órganos con alta tasa de actividad funcional y mitosis. En el presente trabajo evaluamos cambios histológicos en conductos y acinos de glándula submandibular de ratas tratadas con ciclofosfamida, así como el efecto protector de melatonina administrada por vía exógena. Se utilizaron ratas macho Wistar adultas de tres meses de edad con peso corporal de 300 a 350 gramos, divididas en cinco grupos experimentales: 1) C: Control, 2) Control y Etanol, 3) Ciclofosfamida, 4) Ciclofosfamida y Melatonina y 5) Melatonina. Se realizaron cortes histológicos de glándulas submandibulares y tinción con Hematoxilina/Eosina, donde se analizaron posibles cambios estructurales en conductos y acinos. Las glándulas de los animales tratados con ciclofosfamida mostraron células acinares con macrocariosis e hipercromasia, binucleación con contornos irregulares, nucléolo bien evidente y grumos de cromatina perinuclear, respecto de los grupos controles. Se infiere que la administración de melatonina en la dosis ensayada podría atenuar las alteraciones estructurales de la glándula submandibular tratada con este agente quimioterapéutico. La reversión de tales cambios explicaría la reducción de efectos antioxidantes y funcionales a nivel glandular descriptos en nuestro laboratorio. Estudos prévios de nosso grupo demonstraram um efeito oxidativo e funcional desse fármaco, avaliado pelo aumento da superóxido dismutase, do ácido úrico e dos níveis de glicogênio na glândula submandibular de ratos. Por outro lado, a ação da melatonina é conhecida como agente multipropósito por sua capacidade de atenuar alterações na estrutura e função de vários órgãos com alta taxa de atividade funcional e mitose. No presente estudo, avaliamos alterações histológicas nos ductos e ácinos da glândula submandibular de ratos tratados com ciclofosfamida, bem como o efeito protetor da melatonina administrada por via exógena. Utilizaram-se ratos Wistar machos adultos com três meses de idade e peso corporal de 300 a 350 gramas, divididos em cinco grupos experimentais: 1) C: Controle, 2) Controle e Etanol, 3) Ciclofosfamida; 4) Ciclofosfamida e Melatonina e 5) Melatonina. Foram realizados cortes histológicos das glândulas submandibulares com coloração Hematoxilina/Eosina, analisando-se possíveis alterações estruturais em ductos e ácinos. As glândulas dos animais tratados com ciclofosfamida apresentaram células acinares com macrocariose e hipercromasia, binucleação com contornos irregulares, nucléolo proeminente e grumos de cromatina perinuclear, quando comparadas aos grupos controle. Infere-se que a administração de melatonina na dose testada poderia atenuar as alterações estruturais da glândula submandibular tratada com este agente quimioterapêutico. A reversão de tais alterações explicaria a redução dos efeitos antioxidantes e funcionais em nível glandular descritos por nosso laboratório.
