Several reports have described the multipurposeaction of melatonin as an antioxidant agent.This study evaluated superoxide dismutase levels andoxidative stress by quantifying lipid and aqueous peroxidesin an animal model of induced OM treated with theantineoplastic drug fluorouracil and its biomodulator,calcium leucovorin, as well as with melatonin administeredlocally and systemically.Five experimental groups (n=50) were established: G1:Control; G2: fluorouracil/calcium leucovorin; G3: fluorouracil/calcium leucovorin and induced OM; G4: fluorouracil/calcium leucovorin, induced OM, and topicalmelatonin; G5: fluorouracil/calcium leucovorin, inducedOM, and systemic melatonin. Superoxide dismutaseactivity in G3 was lower compared to G4 and G5. Lipidand aqueous peroxide concentrations were higherin G3 than in all treatment groups. Oxidative damagecaused by the deleterious action of the cytostatic agentwas identified in these tissues, alongside the protectivecapacity of melatonin. The exogenous administration ofmelatonin at the tested dose reversed the redox state ofthe buccal mucosa.

Afectalos tejidos blandos de la mucosa oral provocando retrasosen el tratamiento oncológico y alteración dela calidad de vida del paciente. Investigaciones clínicasy experimentales intentaron dilucidar su etiopatogeniacomo así también diversos protocolos farmacológicospara minimizar sus efectos deletéreosen la cavidad oral. Varios reportes describieron laacción de melatonina como agente multipropósitocon poder antioxidante. En el presente trabajo seevaluaron los niveles de superóxido dismutasa y deestrés oxidativo cuantificando peróxidos lipídicos yacuosos, en un modelo animal con MO inducida alque se aplicó la droga antineoplásica fluorouraciloy su biomodulador, la leucovorina cálcica y melatoninapor vía local y sistémica. Se establecieron cincogrupos experimentales (n:50) G1: Control, G2: fluorouracilo/leuovorina cálcica, (G3): fluorouracilo /leucovorina cálcica y MO inducida, Grupo 4 (G4):fluoruracilo / leucovorina cálcica, MO inducida ymelatonina tópica, Grupo 5 (G5): fluoruracilo / leucovorinacálcica, MO inducida y melatonina sistémica.La actividad de superóxido dismutasa en G3fue más baja con respecto a los grupos G4 y G5. Laconcentración de peróxidos lipídicos y acuosos fuesuperior en G3 en relación con todos los grupos detratamiento. Se identificó en estos tejidos el dañooxidativo por la acción deletérea del citostático yla capacidad protectora de melatonina. La administraciónexógena de melatonina en la dosis ensayadarevirtió el estado redox de la mucosa yugal. A mucosite oral (MO) é uma complicação aguda emdoentes submetidos a quimioterapia. Afecta os tecidosmoles da mucosa oral causando atrasos no tratamentodo cancro e alteração da qualidade de vida do doente.A pesquisa clínica e experimental tem tentado elucidara sua etiopatogénese, bem como desenvolver váriosprotocolos farmacológicos para minimizar seus efeitosdeletérios na cavidade oral. Vários relatos descreverama ação da melatonina como um agente polivalente compropriedades antioxidantes. No presente estudo, foramavaliados os níveis de superóxido dismutase e de stressoxidativo, através da quantificação de peróxidos lipídicose aquosos, num modelo animal com OM induzida,no qual foram aplicados local e sistemicamente o fármacoantineoplásico fluorouracil e o seu biomodulador, leucovorinacálcica, além de melatonina administrada porvia local e sistêmica. Foram estabelecidos cinco gruposexperimentais (n:50): G1 Controle, G2 Fluorouracila/leucovorina cálcica, G3 Fluorouracila/leucovorina cálcicae MO induzida, G4 Fluorouracila/leucovorina cálcica,MO induzida e melatonina tópica, G5 Fluorouracila/leucovorina cálcica, MO induzida e melatonina sistêmica.A atividade da superóxido dismutase no G3 foi inferiorà dos grupos G4 e G5. A concentração de peróxidoslipídicos e aquosos foi maior no G3 em relação a todosos grupos de tratamento. Foram identificados nestes tecidosdanos oxidativos causados pela ação deletéria docitostático e a capacidade protetora da melatonina. Aadministração exógena de melatonina na dose testadareverteu, o estado redox da mucosa jugal.
