The gag/pol genomic region plays a fundamental role during the viral\nlife cycle and it has been shown that recombination is a common feature in this region,\nwhich may affect the viral replication capacity (RC) of the recombinant virus. Herein we\nanalyzed the presence of HIV dual infections in individuals with high probability of reinfection\nand we evaluated the impact of intersubtype recombination in the gag/pol\nregion on viral RC and relative RC.\nThree dual infections were detected, two corresponded to individuals co-infected with\nsubtype B and BF recombinant variants, and one was co-infected with two BF\nrecombinant variants. Prolonged infection with a stable clinical condition was observed\nin the three individuals. Resistance mutation patterns were different between the\npredominant and the minority strains. The analysis of Gag and Gagpol sequences\nshowed, for the first time in Argentina, a high natural polymorphism in subtype F and\nBF recombinants variants in comparison with subtype B isolates. The in vitro evaluation\nof RC of two BF recombinant forms revealed that the recombination in the Gag-\nProtease region was associated with a decrease in viral particles production when\ncompared to the B variant. Furthermore, when challenged with subtype B in a dual\ncompetition assay, the BF variants were less efficient than subtype B, reaching a lower\nfrequency in the viral population in a short period of time.\nIn conclusion, our results show that HIV dual infection can occur with closely related\nsubtypes, and even with different variants of the same recombinant form. The\ncomparison of the primary structure of gag/pol sequences between F1 subtype, BF\nrecombinants variants and B subtype isolates, showed that non-B HIV-1 strain were\nhighly polymorphic in positions related with PI exposure and/or resistance. In addition,\nwe describe for the first time the in vitro dynamics of gag-protease-associated RC of\nHIV-1 intersubtype variants, compared with that of subtype B. This study provides\nevidence that intersubtype recombination in this region might alter viral RC generating\nvariants with reduce fitness.\n |

La región gag/pol de HIV-\n1 juega un rol fundamental durante el ciclo de replicación viral, incluyendo ensamblado\ny maduración de la partícula viral. Se ha demostrado que la recombinación intersubtipo\nes frecuente en esta región, lo cual podría afectar la capacidad replicativa de las\nvariantes recombinantes en relación a sus parentales. Teniendo en cuenta lo expuesto\nanteriormente, en el presente trabajo nos propusimos analizar a lo largo del tiempo, la\nvariabilidad genómica del HIV-1 en individuos con infección múltiple y sus posibles\nimplicancias en la dinámica de la población viral y evaluar el impacto de la\nrecombinación intersubtipo en la región genómica gag/pol sobre la capacidad\nreplicativa in vitro en modelos de infección simple y de infección doble.\nEl análisis realizado permitió detectar 3 infecciones dobles, dos de ellas\ncorrespondientes a co-infecciones entre variantes de subtipo B y recombinantes BF, y\nla restante a una co-infección con dos variantes recombinantes BF. La condición\nclínica de los tres individuos se mantuvo estable durante el periodo de estudio y el\npatrón de mutaciones de resistencia entre las variantes mayoritarias y minoritarias fue\ndiferente. Por otra parte, el análisis comparativo de la variabilidad aminoacídica de\nsecuencias de Gag y Proteasa mostró, por primera vez en la Argentina, una mayor\nfrecuencia de cambios polimórficos naturales en los aislamientos de subtipo F y\nvariantes recombinantes BF respecto a los de subtipo B. Por último, la evaluación in\nvitro de la capacidad replicativa de dos variantes recombinantes BF reveló que la\nrecombinación en la región Gag-Proteasa estuvo asociada a una disminución en la\nproducción de partículas virales, respecto a su contraparte de subtipo B. Esta\nobservación fue probada en el contexto de un ensayo de competición entre las\nvariantes mencionadas. Los resultados mostraron que la replicación de las variantes\nrecombinantes fue menos eficiente que su contraparte de subtipo B, alcanzando una\nfrecuencia menor en la población viral en un periodo corto de tiempo.\nNuestros resultados sugieren que la sobreinfección es un fenómeno frecuente en\npoblaciones con riesgo múltiple, aún entre variantes muy relacionadas. El estudio de la\nestructura primaria de secuencias de gag/pol provenientes de aislamientos de\nvariantes recombinantes BF, subtipo F y subtipo B mostró que, las dos primeras\npresentaban mayor cantidad de sustituciones en sitios relacionados con exposición y/o\nresistencia a IP.
