In these circumstances, the atresia process for apoptosis of the granulosa cells (CG), it could be related with the loss of this cellular contacts. In this work, the objective was to establish the relationship among the loss of the contacts Ca++ dependent with the integrity and the apoptosis of the CG.\nConsidering the importance of cadherin (E-CAM and N-CAM) in the development and follicular atresia, which most of the follicles begin to degenerate in the early antral stage and considering the atresia as a stage-dependent process was raised identify and localize the expression of these molecules in the ovary of adult pig, trying to discriminate and relate the different stages of follicle genesis. En antral follicles, those cells with an apparent loss of integrity (lack of cohesion with neighboring cells or disaggregated antral cell layer), did not identify any other label for antibodies in question.\nIn order to correlate the same process in vitro, the study model was a cellular primary culture of porcine CG obtained by aspiration of antrals follicles, which were treated with EGTA, to obtain the disruptión of the homophilic juntions. The treatments were made with EGTA 9 nM and 100 nM and without EGTA, both treatment at different times of induction 6, 12 and 24 hours.\nIn each case, the apoptosis index (AI) was determined by DAPI and TUNEL techniques. Also, was carried out a quantitative evaluation by analysis of images of the expression of Bax by inmuno cytochemical. To different concentration of the inductor, the treatments with serum demonstrated a bigger AI to 9 nM (1,27; 2,58 and 1,38 to 6, 12 and 24 hs respectively). The activation was signifi cant in the treatments of 12 hs with serum (p?0,05) respect to the control. To concentrations of 100 nM the AI didn?t demonstrate signifi cant differences of activation. The quantitative determination of the expression of Bax and for ICQ validates the previous results.\n

Fiorito, C.D. 2010 La fisiología normal de los tejidos reproductivos dependería de un apropiado contacto célula-célula mediado por caderinas, moléculas Ca++ dependientes. El proceso de la atresia dado por apoptosis de las células de granulosa (CG), podría estar relacionado con la pérdida de los contactos celulares. El objetivo del trabajo fue establecer la relación entre la disrupción de estas uniones y la apoptosis de las CG.\nConsiderando la importancia de las caderinas (E-CAM y N-CAM) en el desarrollo y en la atresia folicular, que la mayoría de los folículos empiezan a degenerar en el estadio antral temprano y considerando la atresia como un proceso estadio-dependiente se planteó identifi car y localizar la expresión de estas moléculas en el ovario de cerdo adulto, tratando de discriminar y relacionarla con los distintos estadios de la folículo génesis. En los folículos antrales, aquellas células que presentan una aparente pérdida de integridad (falta de cohesión con células vecinas o desagregadas de la capa de células antrales), no se identifi có marcación alguna para los anticuerpos en cuestión.\nCon la fi nalidad de correlacionar el mismo proceso in vitro, se realizaron cultivos primarios de CG de ovarios porcinos obtenidos por punción en folículos antrales. Los cultivos fueron expuestos por 6, 12 y 24 hs a diferentes concentraciones del inductor de apoptosis, concentraciones de 100 y 9 nM de EGTA (quelante exclusivo de Ca++). Se determinó índice relativo de apoptosis (IRA) mediante la cuantifi cación de las células teñidas con DAPI, TUNEL e inmunocitoquímica (ICQ) con anticuerpos anti Bax. Del mismo modo se determinó el porcentaje de viabilidad por exclusión con Azul Tripán. Se observaron manifestaciones evidentes de apoptosis en los diferentes tratamientos con EGTA, sin embargo las diferencias entre el porcentaje de viabilidad y los IRA por DAPI no son signifi cativas respecto del control.
