TGF-? has been previously shown to exert pro-neurogenic effects on the NPCs present in the hippocampal and subventricular zones (SVZ) and to interact with Notch signaling in different cell types. For this reason, the goal of this work was to study the effect of TGF-? on the commitment of NPCs of the SVZ to a glial fate and its interaction with the Notch signaling pathway.\nInitial cell characterization of NPCs from the SVZ revealed a large percentage of Olig2+, Nestin+ and GFAP+ cell populations, while a small percentage of PDGFR?+ and NG2+ and less than 1% TujI+ cells. Immunocytochemical analysis showed a significant increase in the percentage of PDGFR?+, NG2+ and GFAP+ cells during treatment with TGF-? for 4 days, which is consistent with the increase observed in PDGFR?+ cell proliferation after 24-hour treatment with TGF-?. These results demonstrate the pro-gliogenic effect of this cytokine on the population of NPCs from the adult SVZ. Real-time polymerase chain reaction (PCR) analysis showed TGF-? to induce the expression of Notch ligand and Notch pathway activation target gene Hes1. These findings demonstrate the involvement of Notch signaling in the effects of TGF-?. The characterization of cultures subjected to treatment with TGF-? for 8 days revealed a still high proportion of PDGFR?+ cells and an increase in MBP+ cells. Altogether, the results obtained illustrate the impact of TGF-? on cell fate decisions of NPCs from the SVZ toward a glial cell type, as well as on the proliferation and maturation of oligodendrocyte precursor cells (OPCs). |

Se sabe que las vías de señalización de Notch y el factor de crecimiento transformante ?1 (TGF-?) cumplen un papel crítico en la decisión del destino celular. Previamente se ha demostrado que el TGF-? ejerce un efecto pro-neurogénico en las poblaciones de NPCs presentes en la zona subgranular del giro dentado del hipocampo y en la zona subventricular (SVZ) y es capaz de interactuar con la vía de señalización del receptor Notch en diferentes tipos celulares. Por esta razón, el objetivo del presente trabajo fue estudiar el efecto del TGF-? en el compromiso de las NPC de la SVZ en el destino glial y su interacción con la vía de señalización de Notch.\nLa caracterización celular inicial de cultivos de NPCs provenientes de la SVZ reveló la presencia de una proporción importante de células Olig2+, Nestina+ y GFAP+, mientras que la proporción de células PDGFR?+ o NG2+ fue inferior. Por otro lado se observó menos del 1% de células TujI+. El tratamiento de los cultivos con TGF-? durante 4 días produjo un incremento significativo en del porcentaje de células PDGFR?+, NG2+ y GFAP+ de acuerdo con el aumento en la proliferación de las células PDGFR?+ observado luego de 24hs de tratamiento con TGF-?. Concomitantemente observamos que el TGF-? indujo la expresión de Jagged1, ligando de Notch, y del gen blanco de la activación de esta vía de señalización, Hes1. El tratamiento de los cultivos celulares con TGF-? durante un período más largo mostró que no sólo se mantenía aumentaba la población de células PDGFR?+ sino que indujo un aumento de la población de oligodendrocitos maduros MBP+.
