description The overall aim of this thesis was to evaluate the effect of erythropoietin (EPO) and kit ligand (KITL) during in vitro maturation (IVM) of oocytes.\nTo study the role of both factors during IVM of oocytes we used bovine and murine models. In cattle the effect of adding EPO and KITL to maturation media in the presence of fetal calf serum (FCS) or bovine serumalbumin (BSA) in nuclear and cytoplasmic maturation, the production of reactive oxygen species (ROS) in oocytes and the presence of apoptosis in cumulus cells was studied. In murine the action of both factors during IVM of oocytes obtained from follicles developed in vitro was evaluated.\nAlthough no significant difference were observed in the percentage of nuclear maturation, the number of cleaved bovine embryos and blastocyst obtained increased or tended to increase respect to the control with BSA with the addition of EPO or KITL, respectively. With both factors the number of blastomeres per embryo on day 2 of development was higher, the percentage of cumulus cells undergoing apoptosis decreased and ROS production per embryo was increased compared with the control with BSA.\nIn mouse, the presence of KITL during in vitro maturation of oocytes arising from preantral follicle culture increased the percentage of blastocysts obtained. EPO showed no significant differences.\nIn conclusion, hematopoietic growth factors may have no impact on nuclear maturation of bovine or murine oocytes, however they could have a positive effect on cytoplasmic maturation and developmental competence at least in the conditions studied here.
