Three bacteriological methods can be used, namely: 1° direct examination, 2° culture, 3° inoculation of animals and, in addition to these, serological methods can be added: 4° agglutination, 5° complement fixation. Direct examination is useful for some bacteria, which, by their characteristic appearance in the preparation or by their specific coloration, are distinguished from the countless saprophytes that are often found in preparations made with organic material in non-sterile conditions.
