Although cellular mechanisms are not completely elucidated, a possible cause for this association is the intercellular communication mediated by extracellular vesicles (EVs). Our objective was to evaluate in vitro the role of F and AP in the EVs released by cancer-associated fibroblasts (CAFs). In this sense, CAFs from human mammary adenoma of the F88 line were cultured and stimulated with 40mM F, 250uM AP, F+AP combinations or vehicles for 24h (characteristic concentrations of PD). Supernatants were collected and EVs isolated by sequential ultracentrifugation (2k, 10k, 100k). EVs were observed by transmission electron microscopy for characterization (size and shape) and processed for immunostaining of CD63 EVs marker using colloidal gold, showing a larger number of EVs in F+AP, especially in the 20-30nm particle range; without the treatments altering the EVs shape. In order to evaluate the effect of CAF EVs on cell proliferation, MCF-7 breast epithelial tumor cells were cultured in DMEM 10% FBS and stimulated for 24h with conditioned media and/or with EVs from F88 cells with the different stimuli. MCF7 treated with both conditioned media and EVs from F+AP increased cell proliferation, determined by incorporation of bromodeoxyuridine and cell count (ANOVA, p<0.05). These results suggest a pathogenic effect of food rich in F and AP on breast cancer tumoral microenvironment, through the release of EVs by CAFs, which induce cell proliferation.

Evidencias señalan una mayor adherencia a patrones dietarios (PD) ricos en carbohidratos y ácidos grasos saturados (representados principalmente por fructosa(F) y ácido palmítico(AP)) con el riesgo de desarrollar cáncer de mama. Sin embargo, los mecanismos celulares no están dilucidados completamente, siendo la comunicación intercelular mediada por vesículas extracelulares(EVs) una posible responsable de dicha asociación. Nuestro objetivo fue evaluar in vitro el rol de F y AP en la liberación de EVs por fibroblastos asociados a cáncer(CAFs). Para ello, CAFs de adenoma mamario humano de la línea F88 fueron cultivadas y estimuladas con F 40mM, AP 250uM, combinaciones F+AP o vehículos por 24h (concentraciones características de PD). Se colectaron sobrenadantes y aislaron EVs mediante ultracentrifugaciones secuenciales (2k, 10k, 100k). Las EVs fueron observadas por microscopía electrónica para caracterización (tamaño y forma) y procesadas para la inmunomarcación del marcador de EVs CD63 con oro coloidal, viéndose un mayor número de EVs en F+AP, especialmente en el rango de partículas de 20-30nm; sin que los tratamientos alteren la forma de las EVs. Para evaluar el efecto de las EVs de CAFs sobre la proliferación, células tumorales epiteliales de mama MCF-7 fueron cultivadas en DMEM 10% SFB y estimuladas por 24h con medios condicionados y/o con EVs provenientes de células F88 con los diferentes estímulos. MCF7 tratadas tanto con medios condicionados como con EVs provenientes de F+AP incrementaron la proliferación celular, determinada mediante incorporación de bromodeoxiuridina y conteo celular  (ANOVA, p<0,05). Estos resultados sugieren un efecto patogénico de alimentos ricos en F y AP sobre el microambiente tumoral del cáncer de mama, mediante la liberación de EVs por parte de CAFs, que inducen proliferación celular.
