Affective disorders, including anxiety and depression, are nowadays part of the main threats in public health. Among the most commonly used drugs for the treatment of depression is fluoxetine. However, there are controversial results regarding its side effects, since some authors have shown that these drugs induce a deleterious effect on memory, while others suggest that they improve cognitive function.
The objective of this study was to assess the effects of chronic administration of fluoxetine (10 mg / kg / day) on memory formation processes in Swiss albino mice (N: NIH) with bilateral olfactory bulbectomy compared to mice without extraction of olfactory bulbs (Sham, controls), N = 10 - 14. Memory performance was assessed on day 27 of treatment in the object recognition test (ORT). Subsequently, the level of nitric oxide (NO) in the hippocampus was quantified by Griess technique. In addition, the antidepressant effect in the open field tests (TCA) and tail suspension (TSC) was evaluated. The data were analyzed by two-way ANOVA followed by an LSD test.
The results show that chronic treatment with fluoxetine in Sham animals decreased the percentage of exploration time of the new object in ORT, which is interpreted as a memory deficit (p≤0.05). A similar deleterious effect was observed in bulbectomized animals (BO) administered with saline solution, which was reversed by chronic fluoxetine treatment (p≤0.05). Sham animals that received fluoxetine showed lower concentrations of NO than Sham-saline animals (p≤0.05), while BO-fluoxetine animals had NO concentrations almost double compared to BO-saline (p≤ 0.05). The antidepressant managed to reverse the immobility behavior shown by BO animals in the TSC and hyperlocomotion in TCA (p≤0.05).
Finally, the present work provides new evidence about the effects of fluoxetine on the processes of memory formation in an animal model of depression, indicating that the effects are dependent on the baseline condition of the animal |

Entre los fármacos más utilizados para el tratamiento de la depresión se encuentra fluoxetina, sin embargo existen resultados controversiales en relación a sus efectos secundarios, ya que algunos autores han demostrado que esta inducen un efecto deletéreo de la memoria, mientras otros apuntan a que mejora la función cognitiva. El objetivo del presente trabajo fue evaluar los efectos de la administración crónica de fluoxetina (10 mg/Kg/día) sobre los procesos de formación de la memoria en ratones albino Swiss (N:NIH) con bulbectomía olfatoria bilateral en comparación con ratones sin extracción de los bulbos olfatorios (Sham, controles), N = 10 - 14/grupo. La performance de la memoria fue evaluada en el día 27 de tratamiento en el test de reconocimiento de objetos (TRO). Posteriormente se cuantificó el nivel de óxido nítrico (NO) en hipocampo por técnica de Griess. Además, se evaluó el efecto antidepresivo en los test de campo abierto (TCA) y suspensión de la cola (TSC). Los datos fueron analizados por ANOVA de dos vías seguido por un test de LSD. Los resultados muestran que el tratamiento crónico con fluoxetina disminuyó el porcentaje de tiempo de exploración del objeto nuevo en TRO en animales Sham, lo que se interpreta como un déficit de memoria (p≤0,05). Similar efecto deletéreo fue observado en los animales bulbectomizados (BO) administrados con solución salina, lo cual fue revertido por el tratamiento crónico con fluoxetina (p≤0,05). Los animales Sham que recibieron fluoxetina mostraron concentraciones menores de NO que los animales Sham-salina (p≤0,05), mientras que los animales BO-fluoxetina presentaron concentraciones de NO de casi el doble en comparación con los BO-salina (p≤0,05). El antidepresivo logró revertir la conducta de inmovilidad que mostraron los animales BO en el TSC y la hiperlocomoción en TCA (p≤0,05). En conclusión, el presente trabajo aporta nuevas evidencias acerca de los efectos de fluoxetina sobre los procesos de formación de memoria en un modelo animal de depresión, indicando que los efectos son dependientes de la condición basal del animal Universidad Nacional Cba.
